چکیده

The platelet integrin, α IIb β $ (GPIIb-IIIa), and the tetraspanin, CD9, are integral membrane proteins that are abundant in platelet membranes. We have identified several proteins, including CD9, which were co-precipitated by anti-α IIb β $ antibody from untreated, resting platelets that were solubilized with the poly(oxyethylene) non-ionic detergent, Brij-35. Immunoblot and quantitative immunoprecipitation showed that the association of α IIb β $ with CD9 is specific and stoichiometric. The interaction between CD9 and α IIb β $ is probably hydrophobic, as Triton X100 and hydrophobic detergents of the Brij series completely

Efficient surface expression of platelet GPIIb-IIIa requires both subunits.

Platelet membrane GPIIb-IIIa is a member of the integrin family of heterodimeric adhesion receptors. Processing and export of certain leukocyte and melanoma integrins is disrupted in cells lacking one subunit. We found that surface expression of GPIIb-IIIa, measured by fluorescent activated cell sorting or by surface labeling, required cotransfection of both subunits. In contrast, surface expre...

50-kD integrin-associated protein does not detectably influence several functions of glycoprotein IIb-IIIa complex in human platelets.

A 50-kD integrin-associated protein (IAP) has been reported to be associated with beta 3 integrins and to modulate their function, especially vitronectin receptor in human erythroleukemia (HEL) cells and leukocyte response integrin in neutrophils. We studied the involvement of IAP in the function of platelet beta 3 integrin, glycoprotein (GP) IIb-IIIa complex. IAP was a widely distributed prote...

GPIIb/IIIa inhibitors: from bench to bedside and back to bench again.

From the discovery of the platelet glycoprotein (GP) IIb/IIIa and identification of its central role in haemostasis, the integrin GPIIb/IIIa (αIIbβ3, CD41/CD61) was destined to be an anti-thrombotic target. The subsequent successful development of intravenous ligand-mimetic inhibitors occurred during a time of limited understanding of integrin physiology. Although efficient inhibitors of ligand...

PURIFICATION AND CHARACTERIZATION OF THE HUMAN PLATELET FIBRINOGEN RECEPTOR, GpIIb/IIIa COMPLEX

Platelet GpIIb/IIIa is a member of the integrin family of structurally related adhesion receptors, and adhesive proteins, such as fibrinogen. Ligand binding requires platelet activation by an agonist to convert the receptor to a state where it is capable to bind the ligand. In addition to this a modification, induction of the surface expression and ligand-binding function of an additional pool ...
